Properties:
Pioglitazone is a hypoglycemic drug used to treat type II diabetes mellitus. Belongs to the class of thiazolidinediones. It is a potent and highly selective peroxisomal proliferator-activated gamma receptor (PPAGR) agonist. Activation of nuclear receptors PPAGR modulates the transcription of a number of insulin-sensitive genes that are involved in glucose control and lipid metabolism. Glimepiride reduces the concentration of glucose in the blood, mainly by stimulating the release of insulin from pancreatic beta cells. Its effect is predominantly associated with an improvement in the ability of pancreatic beta cells to respond to physiological glucose stimulation.
Indications:
Treatment of type 2 diabetes mellitus: as a second-line drug if diet, exercise and previous therapy with one or two drugs do not provide adequate control of blood sugar levels.
Dosage and administration:
The recommended dose of the drug is one tablet once a day or as directed by a physician. Dosage and duration of treatment are determined by glycemic control and tolerability.
Contraindications:
Hypersensitivity to pioglitazone, glimepiride or any other component of the drug. Renal failure, sepsis.
Warnings and precautions:
Pioglitazone, like other thiazolidinediones, may cause fluid retention when used alone or in combination with other antidiabetic agents. Patients with CHF require separate pharmacological control. The introduction of sulfonylurea leads to increased mortality due to cardiovascular diseases. Pregnant and lactating women should consult a doctor before using this medicine.
Side effects:
Gastrointestinal disorders, nausea, vomiting, diarrhea, stomach pain, constipation, and a metallic taste in the mouth. Symptoms of side effects, if any, should disappear after the medication is stopped. If symptoms persist, consult your doctor.
