Structure:

Each tablet contains:

Amlodipine 5 mg and valsartan 80 mg

Auxiliary components:

microcrystalline cellulose – 76.76 mg, croscarmellose sodium – 7 mg, povidone K25 – 5 mg, colloidal silicon dioxide – 2.7 mg, magnesium stearate – 1.6 mg.

Properties:

Combined antihypertensive agent. Amlodipine is a third generation BMCC that inhibits the transmembrane entry of Ca2 + into cardiomyocytes and vascular smooth muscle cells, relaxing them. Reduces OPSS, which helps to reduce blood pressure (lying and standing). Heart rate and concentration of catecholamines do not change significantly with prolonged use. The glomerular filtration rate and effective renal plasma blood flow increase without changing the filtration capacity of the kidneys and proteinuria.

Does not inhibit ACE, does not contribute to the accumulation of bradykinin or substance P, therefore, the development of dry cough is unlikely. Does not interact and does not block other hormone receptors or ion channels, which are important for the regulation of the functions of the CVS. A decrease in blood pressure is not accompanied by a change in heart rate. The combination of the two drugs enhances the hypotensive effect compared to that during monotherapy with each drug.

Indications:

Arterial hypertension.

Method of administration and dosage:

The drug should be taken orally with a small amount of water, 1 time / day, regardless of the meal time. The recommended daily dose is 1 tab. drug Amlodipine + Valsartan containing amlodipine / valsartan at a dose of 5/80 mg, 5/160 mg, 10/160 mg. The recommended maximum daily dose is 10/160 mg.

It is recommended to start taking the drug with a dose of 5/80 mg 1 time / day. The dose can be increased after 1-2 weeks from the start of therapy.

Contraindications:

– hypersensitivity to amlodipine, other dihydropyridine derivatives, valsartan, and other auxiliary components of the drug;

– hereditary angioedema, or edema in patients on the background of previous therapy with angiotensin II receptor antagonists;

– severe hepatic impairment (> 9 points on the Child-Pugh scale), biliary cirrhosis, cholestasis;

-severe renal dysfunction (CC less than 30 ml / min);

– use in patients on hemodialysis;

-pregnancy, use in planning pregnancy;

-the period of breastfeeding;

– severe arterial hypotension (systolic blood pressure less than 90 mm Hg);

– age up to 18 years (efficacy and safety have not been established);

-shock (including cardiogenic), collapse;

obstruction of the exit tract of the left ventricle (including hypertrophic obstructive cardiomyopathy (GOKMP)), severe aortic stenosis;

– hemodynamically unstable heart failure after acute myocardial infarction;

– primary hyperaldosteronism;

– simultaneous use with aliskiren in patients with diabetes mellitus or impaired renal function (CC less than 60 ml / min);

Side effects:

From the side of the blood and lymphatic system: very rarely – leukopenia, thrombocytopenia.

From the immune system: very rarely – allergic reactions.

From the side of metabolism and nutrition: very rarely – hyperglycemia.

Mental disorders: infrequently – insomnia, mood lability.

From the nervous system: infrequently – taste disturbances, hypesthesia, tremors, dysgeusia; very rarely – muscle hypertonicity, peripheral neuropathy.

From the side of the organ of vision: infrequently – diplopia.

On the part of the organ of hearing and labyrinthine disorders: infrequently – tinnitus.

From the side of the cardiovascular system: very rarely – arrhythmias (including bradycardia, ventricular tachycardia, atrial fibrillation), myocardial infarction, vasculitis.

From the side of the kidneys and urinary tract: the frequency is unknown – an increase in the concentration of creatinine in the blood plasma, impaired renal function, including acute renal failure.

General disorders: infrequently – increased fatigue, asthenia.
